Understanding Peptide Purity Standards

What HPLC reports

High-performance liquid chromatography separates a sample into its components and measures the relative area of each peak. When a certificate lists "99%+ purity by HPLC", it means the main peak accounts for at least 99% of the total detected peak area under that method. It does not, on its own, confirm the identity of the compound.

Why mass spectrometry matters

Mass spectrometry confirms identity by measuring molecular weight. A complete certificate of analysis pairs a chromatographic purity figure with a mass spec result that matches the expected theoretical mass. Purity without identity confirmation is an incomplete record.

Reading a certificate of analysis

A usable certificate names the compound and lot, states the analytical methods used, reports the results with the acceptance criteria, and identifies the laboratory that performed the work with a test date.
