GHK-Cu: Copper Tripeptide Research and Gene Expression

Origin of the molecule

GHK (glycyl-L-histidyl-L-lysine) is a naturally occurring tripeptide first isolated from human plasma. It binds copper(II) with high affinity to form GHK-Cu, the complex that dominates the published research record. Plasma concentrations of GHK are reported to decline with age, which is one reason the peptide recurs in tissue-remodeling literature.

Gene expression findings

Work by Pickart and colleagues, including analyses published in Cosmetics (2015) and reviewed in Oxidative Medicine and Cellular Longevity, describes GHK-Cu modulating the expression of a broad set of genes, with particular attention to antioxidant enzymes and copper homeostasis pathways. Reviews summarize gene-expression profiling in cultured human fibroblasts in which large numbers of transcripts shifted in the presence of the complex.

Matrix and wound-model literature

Separate reviews cover GHK-Cu in wound-healing and matrix-remodeling models, describing reported influence on collagen and glycosaminoglycan synthesis, metalloproteinase and inhibitor balance, and angiogenic signaling in animal and in-vitro systems. These are laboratory findings; they are not statements about outcomes in people.

Laboratory handling

GHK-Cu is supplied lyophilized. The copper complex is light-sensitive and pH-sensitive, so buffer selection matters when designing assays — chelating buffers can strip the copper and change the species under study.
