Tesamorelin: A GHRH Analogue in the Published Record
What tesamorelin is
Tesamorelin is a stabilized analogue of growth hormone-releasing hormone (GHRH). Rather than supplying growth hormone directly, it acts on the GHRH receptor in the pituitary, which is why the literature commonly tracks IGF-I as a downstream marker of pathway engagement.
Landmark controlled trials
The most cited study is a randomized, placebo-controlled trial of 412 participants published in the New England Journal of Medicine (2007), which measured percent change in visceral adipose tissue by computed tomography over 26 weeks, alongside triglycerides, total-to-HDL cholesterol ratio, IGF-I, and glycemic measures. A later randomized controlled trial in the Journal of Clinical Endocrinology & Metabolism (2012) examined metabolic effects in obese participants with reduced growth hormone secretion.
Why researchers cite it
These trials are frequently used as methodological reference points for imaging-based adiposity endpoints and for study designs that pair a secretagogue mechanism with a biomarker of target engagement. The endpoints, populations, and durations are specific to those protocols and do not transfer to other contexts.
